Biotin (Vitamin H) is an important biomolecule that has been widely used in Biotinylation reactions to label antibodies or other proteins of interest. Due to its low molecular weight, biotin usually does not cause any significant changes in protein conformation and biological activity. Biotinylated proteins/antibodies are widely isolated and assayed with streptavidin pull-down assay, affinity chromatography, ELISA and Western blotting etc. Biovision offers a variety of ready-to-use Biotinylation reagents (Cat. # 2346, 2345, 2347, 2322, 2326, 2323). It is often desired to determine the degree of substitution of such biotinylated biomolecules. Biovision’s Biotin Assay Kit is based on the differential binding of Streptavidin to Biotin and a dye, 2-(4-hydroxyazobenzene) benzoic acid (HABA). When HABA is bound to Streptavidin, addition of free biotin or biotinylated biomolecules results in its displacement in the reaction. These changes are manifested in terms of decrease in the overall absorption (OD 500 nm) and can be quantitatively correlated to the amount of biotin present in solution. The kit also provides Biotin as a Standard and Biotinylated BSA as a positive control. This kit can be used to estimate as low as 60 pmol or 15 ng of biotin in solution.

Carrez clarification reagents are used to treat a wide variety of samples intended to be analyzed by enzymatic or other means. The reagents cause precipitation of proteins and fats, elimination of interference due to a number of redox compounds, which can affect assays and eliminates turbidity and emulsions particularly in food testing. The treatment of samples with Carrez Clarification Reagents is simpler, less expensive and takes less time than other methods such as spin filtering. Use of Carrez reagents is not suitable for samples in which enzymatic activities are to be quantified. Most samples collected for analyses of small molecule analytes such as carbohydrates, alcohols, aldehydes and organic acids can be prepared using this reagent system. These reagents must not be used to prepare samples for analytes such as ascorbate, ammonia, citrate or analytes, which may be converted to these in enzyme based assays, such as urea ( ammonia), aconitate ( citrate). The procedure is quick, simple and dilutes samples by a minimal amount.

β-Lactamase (βL), (EC 3.5.2.6), is a bacterial enzyme that hydrolyzes the β-Lactam ring, a four-carbon ring, the building block of β-Lactam antibiotics. β-Lactams inhibit transpeptidases, enzymes that participate in the biosynthesis of bacterial cell walls. Penams, Carbapenems, Cephems, Clavams, Oxacephems and Monobactams have been widely used for the treatment of bacterial infections since the discovery of penicillin, the first β-Lactam antibiotic, and its bactericidal effect more than eighty years ago. However, an alarming number of cases of β-Lactam antibiotic-resistant infections have been reported. The resistance is found in bacterial strains producing β-Lactamase, which is responsible for diminishing the β-Lactam antibiotic potency. β-Lactamase inhibition, therefore, has become an urgent target in the treatment of bacterial infections displaying β-Lactam resistance. Several β-Lactam derivatives have been reported as β-Lactamase inhibitors; however, only clavulanic acid, sulbactam and tazobactam have reached clinical importance. Beta-Lactamases (βLs), are a large family of hydrolases comprising more than 850 identified members expressed in Gram-positive and Gram-negative bacteria. βLs can be classified according to their substrate or inhibitor specificity. These enzymes are capable of hydrolyzing four atom rings known as β-lactams. Antibiotics containing β-lactam rings (i.e. penicillin, cephalosporin, monobactam, carbapenem) are highly susceptible to be hydrolyzed via enzymatic activity, which deactivates their antibiotic potency. βLs have become a significant clinical threat due to the alarming number of cases of bacterial strains showing β-lactam antibiotic resistance. BioVision’s Beta-Lactamase Activity Assay Kit offers a simple and sensitive assay that can detect and quantify the enzymatic activity of these hydrolases. The Matrix metalloproteinase-1 (MMP-1, Interstitial collagenase, fibroblast collagenase) is a member of a multigene family of calcium-dependent, zinc-containing endoproteinases, the matrix metalloproteinases (MMPs). The MMPs are responsible for the degradation of the extracellular matrix (ECM) including collagens, elastins, gelatin, matrix glycoproteins and proteoglycan during normal development and disease processes. MMPs are regulated by hormones, growth factors and cytokines. MMP-1 belongs to the subclass, the collagenases, and along with MMP-8, and MMP-13 are the only members of the MMP family that are capable of degrading the types I, II and III interstitial collagens with high efficiency. These collagens are primarily found in bone, cartilage and skin. In BioVision’s MMP-1 Inhibitor Screening Kit, MMP-1 hydrolyzes a specific FRET substrate to release a quenched fluorescent group, which can be detected at Ex/Em = 490/520nm. In presence of potent MMP-1 inhibitors the clevage of substrate will be inhibited or stopped. The kit provides a rapid, simple, sensitive, and reliable test suitable as a high throughput screening assay of MMP-1 inhibitors. Firefly luciferase has been used as a sensitive reporter for studying gene regulation and function. Luciferase provides a 1000-fold increase in sensitivity in comparison to the standard chloramphenicol acetyltransferase (CAT) assay. Firefly luciferase catalyzes the oxidative carboxylation of luciferin, a reaction with the highest efficiency of any known bioluminescence reaction. The light emission from the reaction can be recorded using a luminometer. BioVision’s Luciferase Reporter Assay Kit provides a simple means for detecting luciferase activity in transfected eukaryotic cells. For measurement of expressed luciferase in vitro, luciferase is first extracted from transfected cells through cell lysis. Then CoA, ATP, Mg²ᶧ and buffer are added to the lysate (Inclusion of CoA yields a nearly constant light emission rather than typical flash Kinetics). The luminescent reaction is then triggered by an injection of luciferin, and the emitted light is recorded by a luminometer. By providing CoA, along with ATP, Mg²ᶧ in an optimized buffer solution, BioVision’s Luciferase Reporter Assay Kit ensure maximal sensitivity, consistent light output, as well as convenience and consistency when working with multiple samples.

Renin (EC 3.4.23.15), also known as an angiotensinogenase, is an enzyme that participates in the renin-angiotensin system (RAS) which mediates extracellular volume (i.e. blood plasma, lymph and interstitial fluid), and arterial vasoconstriction. An over-active renin-angiotensin system leads to vasoconstriction and retention of sodium and water, causing hypertension. Renin inhibitors are widely used for the treatment of hypertension. In BioVision’s Renin Activity Assay Kit, Renin and other proteases hydrolyze the FRET substrate resulting in a product that is detected fluorometrically at Ex/Em = 328/552 nm to give total protease activity. In the presence of a Renin-Specific Inhibitor, hydrolysis of the substrate is only due to the non-specific protease activity. The difference between the total activity and the activity in the presence of Renin Specific Inhibitor, gives the Renin Activity in the sample. This rapid, simple & sensitive kit can detect renin activity as low as 0.75 U/ml.
