Alanine Colorimetric/Fluorometric Assay Kit » Descripción

Alanine is the 2nd most abundant of the 20 proteinogenic amino acids. Nonessential, being available from dietary sources, it plays a key role in the glucose-alanine cycle between tissues and liver. In muscle and other tissues that degrade amino acids, amino groups are pooled as glutamate by transamination. Glutamate then transfers the amino group to pyruvate via alanine aminotransferase, forming alanine and α-ketoglutarate. The alanine is passed into the blood and transported to the liver. A reverse of the alanine aminotransferase reaction takes place in liver. Pyruvate can be used in gluconeogenesis, to form glucose which may return to other tissues through the circulatory system. There appears to be a correlation between alanine levels and and higher blood pressure, energy intake, cholesterol levels, and body mass index. BioVision’s Alanine Assay Kit provides a sensitive detection method of alanine. In the kit, alanine is converted to pyruvate which is specifically detected leading to proportional color (λ=570nm: 0-10 nmol) or fluorescence (Ex/Em 535/587nm: 0-1 nmol) development. Serum concentration: ~24-76 µg/ml (~3-9 nmol/10 µl).

β-Hydroxybutyrate (β-HB) is the most abundant of the ketone bodies (~78% of total ketone bodies in blood). Ketoacidosis (high concentrations of the ketone bodies) can be caused by a variety of conditions, such as diabetes, alcoholism and severe starvation. Ketoacidosis can be fatal if not treated. Additionally, ketogenic diets can increase the concentration of ketone bodies slightly in healthy individuals. BioVision’s β-Hydroxybutyrate Assay Kit offers simplicity, enhanced sensitivity, and can be adapted to high-throughput applications. The assay is based on enzymatic oxidation of β-HB that results in generation of fluorescent signal (Ex/Em = 535/587 nm) which is directly proportional to the amount of β-HB. The kit offers an excellent alternative for measurement of β-HB when biological sample quantities are limited or subketogenic levels are suspected. The assay can detect β-Hydroxybutyrate as low as 4 µM in a variety of biological samples.

Acetoacetate (AcAc), a β-ketoacid, is one of the three ketone bodies and is formed via condensation of two molecules of acetyl-CoA in liver mitochondria. AcAc can be enzymatically reduced to 3-β-hydroxybutyrate (β-HB), or decarboxylated producing acetone (CH3)2CO). Ketone bodies (βHB: 78%; AcAc: 20% & CH3)2CO: 2%) are mainly used as an alternative energy source when glucose cannot be delivered to the system. Excessive concentration of ketone bodies (ketoacidosis) is observed in patients with Type I diabetes, severe starvation or alcoholism. Traditionally, AcAc levels have been qualitatively detected using dipsticks that use sodium nitroferricyanide as a chromophore. BioVision’s Acetoacetate Assay Kit has adapted that principle with a modification that provides a sensitive method to quantitate endogenous levels of AcAc in human blood, and urine. In this non-enzymatic assay, AcAc reacts with a substrate to generate a colored product that can be measured at 550 nm. The reaction is specific for AcAc and does not detect 3-β-hydroxybutyrate. The assay kit can detect samples containing acetoacetate as low as 25 µM.

Carbohydrates play important structural as well as chemical roles in all living systems. Detection of total carbohydrates, therefore, has wide applications. BioVision’s Total Carbohydrate Assay is a simple, sensitive and robust method of detecting virtually all carbohydrates. The assay is based on the phenol-sulfuric acid method. In Total Carbohydrate Assay, polysaccharides (mono, di, tri, etc.) and their derivatives, in the presence of sulfuric acid, are hydrolyzed to monomers and converted to furfural or hydroxyfurfural, which react with the Developer to form a chromogen that can be quantified by measuring the absorbance at 490 nm. The Total Carbohydrate Assay can detect most forms of carbohydrates, including simple and complex saccharides, glycans, glycoproteins and glycolipids.

D-Mannitol is one of the most abundant sugar alcohols in nature. It can be produced in the biosphere by a range of organisms such as bacteria, yeast, fungi and almost all plants. Mannitol functions as a carbohydrate storage depot, as a scavenger for reactive oxygen species and as an osmoprotectant. Recent studies have shown that increased mannitol content in plants protects them against fungal infections and improves resistance to drought and high salinity. The mannitol pathway reversibly converts fructose to mannitol and serves as the main carbohydrate cycle for fungi survival. Measurement of mannitol level is important for evaluating mannitol metabolic pathways and commercially, in developing fungi and drought resistant plants. In BioVision’s D-Mannitol Assay Kit, D-mannitol is converted to D-fructose by mannitol dehydrogenase in the presence of NAD to form NADH, which reduces a colorless probe to a chromogen with strong absorbance at 450 nm.
