Inorganic phosphate (Pi) is one of the most important ions in biological systems. It functions in a variety of roles. One of the most important roles is as a molecular switch, turning enzyme activity on and off through the mediation of the various protein kinases and phosphatases in biological systems. A highly sensitive assay is desired to monitor Pi in variety samples or monitor Pi changes during kinase and phosphatase reactions. The newly designed Phosphate Assay Kit provides a sensitive, easy, quick means of assessing phosphate over a wide range of concentrations. In the assay, inorganic phosphate reacted with maltose to produce glucose in the present of a special enzyme. The glucose is specifically oxidized to generate a product that reacts with the OxiRed® probe to generate fluorescence (Ex/Em=535/587nm). The kit can be used to detect Pi produced through reactions involving ATPases, GTPases, 5’-nucleotidase, protein phosphatases, acid and alkaline phosphatases, and phosphorylase, etc. from a variety of samples. Glucose background can be eliminated by doing a glucose control. Phosphate concentrations between 0.4 µM and 10 µM, with a lower detection limit of approximately 40 pmol can be directly determined.

Inorganic phosphate (Pi) is one of the most important ions in biological systems. It functions in a variety of roles. One of the most important roles is as a molecular switch, turning enzyme activity on and off through the mediation of the various protein kinases and phosphatases in biological systems. The Pi ion is also of great importance in mineralization processes and is a primary stimulus of algal blooms frequently found in bodies of fresh water, due to runoff from areas of high fertilizer use. The PicoProbe Phosphate Assay Kit provides an easy, quick and sensitive means of assessing phosphate over a wide range of concentrations. In the assay, inorganic phosphate will react with substrate and PicoProbe™ to generate fluorescence (Ex/Em = 535/587 nm). The kit can be used to detect Pi in a variety of samples or to monitor phosphate released by an assortment of enzymes (such as ATPases, GTPases, 5’-nucleotidase, protein phosphatases, acid and alkaline phosphatases, and phosphorylase kinase). Unlike other commercially available assays, the PicoProbe assay is not affected by the presence of glucose in samples (Note: Glucose interferes with many other commercially available assays). This PicoProbe assay is highly sensitive with the detecting limit approximately 40 pmol/well.

Adipose tissue secretes a number of biologically active soluble factors (collectively named adipocytokines) that regulate glucose and fatty acid metabolism. Measurement of serum adiponectin levels gives us important information on the role of adiponectin in regulation of glucose and/or lipid metabolism. This Rat Adiponectin ELISA Kit is an enzyme-linked immunosorbent assay (ELISA) for quantitative determination of adiponectin in rat serum, plasma or various tissue or cell culture supernatants. In the assay, polyclonal antibody specific for rat adiponectin has been pre-coated onto 96 well microplate. Standards and samples are pipetted into the wells and any adiponectin present is bound by immobilized antibody. The bound adiponectin is then captured by anti-rat adiponectin monoclonal antibody. With adding HRP conjugated mouse IgG and HRP substrate, the colors developed in proportion to the bound adiponectin, can be easily measured by Elisa plate reader.

Phosphate is also of great importance in mineralization processes and is a primary stimulus of algal blooms frequently found in bodies of fresh water, due to run-off from areas of high fertilizer use. The newly designed Phosphate Colorimetric Assay Kit provides an easy, quick and sensitive means of assessing phosphate over a wide range of concentrations. The assay utilizes a proprietary formulation of malachite green and ammonium molybdate which forms a chromogenic complex with phosphate ion giving an intense absorption band around 650 nm. Phosphate concentrations between 1 µM and 1 mM, with a lower limit of detection of approximately 0.1 nmol, can be directly determined. The Phosphate Colorimetric Assay Kit provides 500 assays using microtiter plates or 100 assays using 1 ml cuvettes.
